What are magnetic DNA bead cleanups?
DNA SPRI bead cleanups are often used after PCR reactions to remove residual primers, nucleotides, enzymes, salts, and other components that may interfere with downstream processing or analysis. SPRI beads are paramagnetic particles used to reversibly bind DNA in the presence of polyethylene glycol and salts. After the DNA binds to the bead surface, a magnet collects the particles against the well wall. The supernatant containing unwanted reaction components is removed, the bead-bound DNA is washed with ethanol, residual ethanol is allowed to vaporate, and the purified DNA is eluted in a low-salt buffer or water for downstream use.
