How does cell seeding impact assay reproducibility?
Reliable cell-based assays depend on the consistent and gentle handling of live cell suspensions. Whether the assay measures viability, proliferation, cytotoxicity, metabolism, receptor activation, or drug response, each well must receive a consistent number of healthy, viable cells. Variability introduced during cell seeding can affect downstream assay signal, increase well-to-well variation, and reduce confidence in experimental results. Liquid handling is an important source of variability in cell-based assay setup. Cell suspensions require careful handling because cells can settle over time, may be sensitive to shear stress, and can respond differently to aspiration and dispensing conditions. Pipetting too quickly may increase mechanical stress on cells, while inconsistent technique can contribute to differences in cell number and viability across wells. In plate-based assays, even small differences between wells can influence data quality and reproducibility.
