Microbial contamination monitoring in pharmaceutical-grade waters has relied on laboratory-based methods developed over a century ago. These are labor-intensive techniques that require lab personnel to take samples manually from sampling ports. The samples are then cultured on a plate to determine if any colonies form that indicate microbial contamination. This method has a number of potential sources of contamination, even when handled by highly-skilled personnel. This type of contamination can result in false positive readings, which then lead to additional testing expenses, costly investigations and, as a worst case, scrapping of the production batch even though it is not truly contaminated.

An alternative to laboratory-based techniques that is gaining wide acceptance from regulators and the industry, is rapid microbial methods (RMMs). These are real-time, on-line technologies for determining bioburden in a sample.
