Determination of Sucrose Content in Apple Juice

How is Sucrose Content Measured Spectrophotometrically?

Sucrose is a disaccharide composed of glucose and fructose. The sucrose Assay kit is used for the quantitative enzymatic determination of sucrose in food and other materials. The sucrose is hydrolyzed to glucose and fructose by the enzyme invertase.

This glucose and fructose is phosphorylated to Glucose - 6 - phosphate (G6P) by ATP (Adenosine triphosphate) with hexokinase as a catalysator. Afterwards, G6P dehydrogenase (G6PDH) catalyzes the reaction, where nicotinamide adenine dinucleotide (NAD) oxidizes the G6P to 6-phosphogluconate. During this oxidation, NAD is reduced to NADH. Because NADH absorbs at 340 nm. The increase in absorbance is directly proportional to the reduced NADH and to the sucrose concentration.
